ABEMACICLIB

Generic Name: ABEMACICLIB

ABSTRACT -

Abemaciclib is an oral CDK4/6 inhibitor used in selected hormone receptor-positive, HER2-negative breast cancer settings. By inhibiting CDK4 and CDK6, it reduces retinoblastoma-protein phosphorylation and slows cell-cycle progression from G1 to S phase.

DOSAGE –

Use the current locally approved prescribing information where a product is authorized. For investigational products, dose, route, schedule, premedication, and dose modifications must come only from the applicable clinical-trial protocol. No dose should be selected from this summary.

SIDE EFFECTS –

  • Infusion- or injection-site reactions
  • Headache or fatigue
  • Fever or chills
  • Rash or itching
  • Infections or blood-count changes, depending on target
  • Target-related immune or organ toxicity
  •  

ALLRGIC REACTIONS–

(These symptoms may indicate a serious allergic or infusion reaction and require medical attention.)

  • Hives or a rapidly spreading rash
  • Swelling of the face, lips, tongue, or throat
  • Wheezing, breathing difficulty, or sudden dizziness
  • Severe blistering, mucosal lesions, or anaphylaxis

DRUG INTERACTIONS - 

  • Strong CYP3A inhibitors may increase exposure
  • Strong CYP3A inducers may reduce exposure
  • Other myelosuppressive medicines can increase cytopenias

PRECAUTIONS –

  • Monitor blood counts and liver function
  • Treat diarrhoea promptly and maintain hydration
  • Assess for thrombosis and new respiratory symptoms
  • Follow pregnancy-prevention guidance

WARNINGS –

  • Severe diarrhoea, neutropenia, hepatotoxicity, thrombosis, and pneumonitis can occur
  • Fever with neutropenia requires urgent assessment
