ANTITHROMBIN ALFA
Generic Name: ANTITHROMBIN ALFA
ABSTRACT -
Antithrombin alfa is recombinant human antithrombin, marketed as ATryn in some regions.
It is used for prevention of peri-operative and peri-partum thromboembolic events in patients with hereditary antithrombin deficiency.
It restores antithrombin activity and enhances physiologic inhibition of thrombin and factor Xa.
DOSAGE -
- Dosage should be determined only by a qualified healthcare professional.
- For intravenous use after reconstitution.
- Dosing is individualized using baseline and target antithrombin activity; therapy commonly uses a loading dose followed by continuous infusion.
- Antithrombin activity should be monitored and the dose adjusted to maintain the target range specified by the label or specialist protocol.
SIDE EFFECTS–
- Possible side effects may include:
- Bleeding
- Hematoma or bruising
- Infusion-site reaction
- Nausea
- Headache
- Hematuria
- Hypersensitivity or anaphylaxis
- Antibody development is possible but uncommon
ALLRGIC REACTIONS–
- Hypersensitivity to the active biologic or formulation excipients is possible.
- Symptoms may include rash, urticaria, pruritus, facial swelling, bronchospasm, fever, chills, hypotension, or anaphylaxis.
- Patients with a suspected allergic reaction should stop treatment and seek urgent medical attention.
DRUG INTERACTIONS–
- Heparin and low-molecular-weight heparins may have enhanced anticoagulant effect when antithrombin is restored.
- Other anticoagulants, antiplatelet drugs, thrombolytics, and NSAIDs can increase bleeding risk.
- Monitor coagulation and bleeding signs when used with anticoagulants.
PREAUTIONS–
- Confirm hereditary antithrombin deficiency and indication before treatment.
- Monitor antithrombin activity and bleeding.
- Use caution in active bleeding or high hemorrhage risk.
- Pregnancy/peripartum use requires specialist obstetric and hematology oversight.
WARNINGS –
- Serious bleeding can occur, especially with concomitant anticoagulants.
- Hypersensitivity reactions can occur with recombinant biologic products.
- Not indicated for treatment of established thromboembolic events unless directed by a specialist.
