ALISPORIVIR
Generic Name: ALISPORIVIR
ABSTRACT -
Alisporivir, also known as Debio 025, is an investigational non-immunosuppressive cyclosporin analogue that inhibits host cyclophilins. It has been studied as an antiviral candidate, particularly for hepatitis C virus, where cyclophilin inhibition can interfere with viral replication. It is not an approved routine therapy and should be used only in clinical-trial or specialist settings.
DOSAGE -
- Alisporivir is investigational and does not have a universally established approved dosage regimen.
- Clinical studies used protocol-defined oral dosing schedules, often in combination with other antiviral agents.
- Dose, duration, food instructions, laboratory monitoring, drug-combination rules, and stopping criteria must follow the study protocol.
- Patients should not use alisporivir for self-medication or as a substitute for approved antiviral therapy.
SIDE EFFECTS–
- Reported or potential side effects may include:
- Nausea
- Diarrhea
- Headache
- Fatigue
- Abdominal discomfort
- Elevated bilirubin
- Elevated liver enzymes
- Anemia or laboratory abnormalities
- Rash
- Pancreatitis-like abdominal pain reported in antiviral-development settings
- Allergic or hypersensitivity reactions
ALLRGIC REACTIONS–
(These symptoms may indicate an allergic reaction to the drug.)
- Skin rash
- Itching
- Hives
- Swelling of face, lips, tongue, or throat
- Difficulty breathing
- Wheezing
- Severe dizziness
- Rapid heartbeat
- Chest tightness
- Anaphylaxis
- Severe hypersensitivity reactions
DRUG INTERACTIONS–
- The interaction profile may include or require caution with:
- Other hepatitis C or antiviral medicines
- Strong CYP3A or P-glycoprotein inhibitors or inducers
- Cyclosporine, tacrolimus, sirolimus, or related immunosuppressants
- Azole antifungals, macrolide antibiotics, certain antivirals, or anticonvulsants
- Statins or other medicines affected by transporter inhibition
- Medicines associated with liver or pancreatic toxicity
- Other investigational medicines
- Patients should inform healthcare providers about all prescription medicines, non-prescription medicines, herbal products, and supplements being used.
PREAUTIONS–
- Use only in a clinical-trial, hepatology, infectious-disease, or specialist-supervised setting.
- Assess baseline liver function, bilirubin, pancreatic enzymes when indicated, renal function, viral status, and interacting medicines.
- Monitor for jaundice, abdominal pain, pancreatitis symptoms, liver laboratory abnormalities, and drug-interaction toxicity.
- Use caution in patients with significant liver disease, pancreatic disease, kidney impairment, or complex antiviral regimens.
- Pregnancy, breastfeeding, and pediatric safety have not been established.
WARNINGS –
- Alisporivir is investigational; safety, efficacy, optimal dose, and routine clinical role have not been fully established.
- Severe abdominal pain, jaundice, dark urine, persistent vomiting, severe fatigue, or signs of pancreatitis requires urgent evaluation.
- Drug interactions may be clinically important because alisporivir is related to cyclosporin-type molecules.
- Do not combine with other antivirals or immunosuppressants unless directed by a specialist protocol.
- Keep out of reach of children.
