CENZILEUCEL
Generic Name: CENZILEUCEL
ABSTRACT -
Cenzileucel is an autologous chimeric antigen receptor T-cell (CAR-T) therapy under development for the treatment of relapsed/refractory haematological malignancies. Patient-derived T cells are engineered ex vivo to express a CAR targeting a tumour-associated antigen, enabling selective recognition and elimination of malignant cells. It is being evaluated in Phase I/II clinical trials in patients with limited therapeutic options.
DOSAGE -
Single intravenous infusion following lymphodepleting chemotherapy conditioning; cell dose defined in units of CAR-T cells per kg body weight, as per clinical protocol.
SIDE EFFECTS-
- Cytokine release syndrome (CRS)
- Immune effector cell-associated neurotoxicity syndrome (ICANS)
- Cytopenias
- Infections
- Fatigue
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Infusion hypersensitivity
- Fever and rigors
- Anaphylaxis (rare)
- Urticaria
DRUG INTERACTIONS-
- Avoid live vaccines for ≥6 months post-infusion
- Corticosteroids used to manage CRS may reduce CAR-T efficacy
- Avoid immunosuppressants that may impair T-cell function
PREAUTONS–
- Patients must be monitored in a certified healthcare facility for at least 7 days post-infusion
- Neurological status assessments required post-infusion
- Long-term follow-up for secondary malignancy required
- B-cell aplasia expected in CD19-targeted products
WARNINGS –
- CRS and ICANS can be life-threatening — tocilizumab and corticosteroids must be available
- Prolonged cytopenias may require supportive care
- Manufactured under strict GMP conditions — product is patient-specific
