ABSTRACT -
Cabamiquine (M5717) is an oral antimalarial agent developed by Merck targeting Plasmodium falciparum elongation factor 2 (eEF2), a protein essential for parasite protein synthesis. It demonstrates potent activity against multiple Plasmodium life-cycle stages including asexual blood stages, liver stages, and gametocytes, potentially blocking transmission. Phase II trials evaluated single-dose or short-course regimens for uncomplicated falciparum malaria.
DOSAGE -
Oral tablet; studied as single-dose or 3-day regimen (e.g., 800 mg loading dose with partner drug) in Phase II trials.
