CAGRILINTIDE
Generic Name: CAGRILINTIDE
ABSTRACT -
Cagrilintide is a long-acting amylin analogue (acylated) co-developed with semaglutide (as CagriSema) by Novo Nordisk for obesity and type 2 diabetes. Amylin acts centrally to promote satiety, reduce food intake, and slow gastric emptying. Its extended half-life allows once-weekly subcutaneous dosing. Phase III trials (REDEFINE programme) demonstrate significant body weight reduction when combined with semaglutide.
DOSAGE -
Subcutaneous injection once weekly; dose escalated from 0.25 mg to 2.4 mg over 16 weeks; used as fixed-ratio combination with semaglutide 2.4 mg (CagriSema).
SIDE EFFECTS -
- Nausea
- Vomiting
- Diarrhoea
- Constipation
- Decreased appetite
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Injection site reactions (erythema, induration)
- Drug hypersensitivity
- Urticaria
- Anaphylaxis (rare)
DRUG INTERACTIONS-
- Additive GI effects with GLP-1 receptor agonists (given as combination)
- May slow gastric emptying — alter oral drug absorption
- Monitor glucose when combining with antidiabetic agents
PREAUTONS–
- Personal or family history of medullary thyroid carcinoma — avoid
- Multiple endocrine neoplasia type 2 (MEN 2) — contraindicated
- Pancreatitis history — use with caution
- Pregnancy and lactation: avoid
WARNINGS –
- Thyroid C-cell tumours in rodents with amylin and GLP-1 analogues — class warning
- Risk of hypoglycaemia when combined with insulin or sulfonylureas
- Gallbladder disease including cholecystitis reported
