CROMAKALIM
Generic Name: CROMAKALIM
ABSTRACT -
Cromakalim is an ATP-sensitive potassium (KATP) channel opener formerly studied for hypertension, gastric ulcer, and cardiomyopathy-related indications. Development was discontinued after heart lesions were observed in monkey chronic-toxicity studies.
DOSAGE -
No approved clinical dosage exists and the API should not be used for routine human treatment. Research dosing, if any, must follow an approved protocol.
SIDE EEFECTS -
Expected pharmacologic effects include hypotension, dizziness, headache, flushing, reflex tachycardia, palpitations, edema/fluid retention, and syncope. Animal cardiac-toxicity findings are a major development concern.
ALLERGIC REACTIONS -
Hypersensitivity is not characterized; rash, urticaria, angioedema, respiratory symptoms, or anaphylaxis should be treated as possible allergy.
DRUG INTERACTIONS-
Additive hypotension may occur with antihypertensives, nitrates, PDE5 inhibitors, diuretics, or alcohol. Sulfonylureas may theoretically oppose KATP-channel opening.
PREAUTONS–
Avoid in patients with hypotension, significant cardiovascular disease, edema/heart failure, or arrhythmia risk outside research. Monitor blood pressure, heart rate, ECG/cardiac markers if studied. Pregnancy/lactation safety is unknown.
WARNINGS –
Development discontinued because of heart lesions in monkey chronic-toxicity studies; no approved human use.
