COTADUTIDE
Generic Name: COTADUTIDE
ABSTRACT -
Cotadutide is a synthetic peptide co-agonist of glucagon-like peptide-1 (GLP-1) and glucagon receptors, designed to promote weight loss through appetite suppression, energy expenditure enhancement, and improved glycemic control. GLP-1 agonism reduces appetite and glucose excursions, while glucagon agonism drives hepatic lipid oxidation and thermogenesis. It is under clinical investigation for obesity, NASH/MASH, and type 2 diabetes.
DOSAGE -
Administered subcutaneously once daily; dose titration regimen under clinical investigation.
SIDE EEFECTS -
- Nausea (common, dose-dependent)
- Vomiting
- Decreased appetite
- Diarrhea
- Hypoglycemia (in diabetic patients)
ALLERGIC REACTIONS -
(These symptoms may indicate an allergic reaction and require medical attention.)
- Injection site reactions
- Urticaria or rash
- Anaphylaxis (rare)
- Angioedema
DRUG INTERACTIONS-
- Concomitant insulin or sulfonylureas — increased hypoglycemia risk
- Oral medications — delayed gastric emptying may alter absorption
- Antidiabetic agents require dose adjustment
PREAUTONS–
- Monitor for pancreatitis (abdominal pain, lipase elevation)
- History of medullary thyroid carcinoma or MEN 2 — contraindicated
- Renal function monitoring in dehydration from GI side effects
- Pregnancy safety not established
WARNINGS –
- Risk of thyroid C-cell tumors based on rodent data (class warning)
- Acute pancreatitis — discontinue if suspected
- Embryo-fetal toxicity — contraception required
