CAMAZEPAM
Generic Name: CAMAZEPAM
ABSTRACT -
Camazepam is a benzodiazepine derivative with anxiolytic, sedative, hypnotic, and muscle relaxant properties, acting as a positive allosteric modulator of GABA-A receptors. It is metabolised to diazepam and other active benzodiazepines. Historically used in several European countries for anxiety disorders and insomnia, it is now rarely prescribed due to concerns about dependence and the availability of safer alternatives.
DOSAGE -
10–20 mg orally, 1–3 times daily for anxiety; dosing adjusted to the minimum effective dose for the shortest possible duration.
SIDE EFFECTS -
- Sedation and drowsiness
- Cognitive impairment
- Ataxia
- Dependence and withdrawal syndrome
- Paradoxical excitation (rare)
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Skin rash
- Urticaria
- Angioedema
- Anaphylaxis (rare)
DRUG INTERACTIONS-
- CNS depressants (alcohol, opioids, antipsychotics) — fatal respiratory depression risk
- CYP3A4 inhibitors increase camazepam/diazepam levels
- Rifampicin and other CYP inducers reduce efficacy
PREAUTONS–
- Use for shortest duration necessary — dependence risk
- Avoid in sleep apnoea, myasthenia gravis, and severe respiratory insufficiency
- Dose reduction required in elderly and hepatic impairment
- Abrupt discontinuation risks seizures — taper gradually
WARNINGS –
- Physical and psychological dependence — risk increases with dose and duration
- Respiratory depression with opioids — co-prescription requires explicit justification
- Contraindicated in pregnancy — neonatal withdrawal and floppy infant syndrome
