AMDOXOVIR

Generic Name: AMDOXOVIR

ABSTRACT -

Amdoxovir (DAPD, AMDX) is an investigational guanosine analogue nucleoside reverse transcriptase inhibitor studied for HIV infection. It is deaminated intracellularly to dioxolane guanine (DXG), which is then phosphorylated to an active triphosphate that inhibits viral reverse transcriptase and can terminate viral DNA synthesis. Development remained investigational; it is not an approved substitute for guideline-based antiretroviral therapy.

This document is for structured API information only. It is not a prescribing guide. Clinical use, if any, must follow the approved local label, trial protocol, and specialist medical supervision.

DOSAGE -

Dosage should be determined only by a qualified healthcare professional.

  • No approved routine dosing regimen is available.
  • Clinical-trial dosing was protocol-specific and used with other antiretroviral agents.
  • Do not start, stop, or substitute HIV medicines without an infectious-disease specialist.

SIDE EFFECTS–

Possible side effects and safety concerns include:

  • Nausea, vomiting, diarrhoea, or abdominal discomfort
  • Headache or dizziness
  • Fatigue or malaise
  • Liver enzyme abnormalities
  • Possible nucleoside-analogue class effects such as pancreatitis, neuropathy, lactic acidosis, or hepatic steatosis
  • Lens opacity/cataract monitoring was raised as a development concern in clinical safety information.

ALLRGIC REACTIONS–

Hypersensitivity to Amdoxovir or related compounds/products may occur. Symptoms may include:

  • Rash or itching
  • Urticaria
  • Facial, lip, or throat swelling
  • Shortness of breath
  • Severe hypersensitivity reaction

Important: Patients who develop an allergic reaction to Amdoxovir should seek immediate medical attention and inform all future healthcare providers of the allergy.

DRUG INTERACTIONS–

Amdoxovir may interact with certain medicines, vaccines, protocols, or clinical conditions. Caution is needed with:

  • Other nucleoside or nucleotide analogues may increase mitochondrial-toxicity risk.
  • Hepatotoxic medicines may increase liver-monitoring requirements.
  • Renally cleared medicines require careful review if renal function changes.
  • Combination antiretroviral regimens must be selected by an HIV specialist.

Patients should inform healthcare providers about all prescription medicines, over-the-counter medicines, herbal products, vaccines, and supplements they are using.

PREAUTIONS–

  • Assess renal function, hepatic function, HIV resistance profile, and complete medication list before investigational use.
  • Monitor for symptoms of lactic acidosis, pancreatitis, neuropathy, ocular changes, and hepatic injury.
  • Use only as part of an effective combination regimen in a clinical protocol.
  • Pregnancy and lactation safety are not established.

WARNINGS –

Amdoxovir is investigational and not approved for routine HIV treatment.

  • Inadequate antiretroviral therapy can cause viral resistance and treatment failure.
  • Nucleoside analogue class toxicities can be serious or life-threatening.
  • Keep out of reach of children.
