ABSTRACT -
Dactolisib is an investigational orally bioavailable inhibitor of phosphoinositide 3-kinase and mTOR signaling. It has been studied in oncology and related signaling-driven diseases.
DOSAGE -
- No routine clinical dosage is established for general use.
- Dose, schedule, fasting/food instructions, and dose modifications must follow clinical-trial protocol.
- Metabolic and hepatic monitoring are important for PI3K/mTOR pathway inhibitors.
SIDE EEFECTS -
- Nausea, vomiting, diarrhea, fatigue, mucositis, rash, decreased appetite, or headache may occur.
- Hyperglycemia, hyperlipidemia, liver enzyme elevation, cytopenias, and infection may occur.
- Pneumonitis-like symptoms are potential class concerns.
ALLERGIC REACTIONS -
- Immediate rash, hives, itching, flushing, or generalized skin reaction
- Facial, lip, tongue, or throat swelling
- Wheezing, chest tightness, shortness of breath, or severe dizziness
- Severe hypotension, collapse, or anaphylaxis requires emergency care
- Any suspected allergy should be documented and reported to future healthcare providers
DRUG INTERACTIONS-
- CYP modulators, antidiabetic therapy, immunosuppressants, and other anticancer agents require review.
- Other hepatotoxic or myelosuppressive agents may increase risk.
- Patients should report all medicines and supplements.
PREAUTONS–
- Monitor glucose, lipids, liver tests, CBC, infections, mucositis, rash, and pulmonary symptoms.
- Use caution in diabetes, infection, hepatic impairment, pregnancy, and lactation.
- Contraception counseling may be needed.
WARNINGS –
- Severe hyperglycemia, pneumonitis, infection, mucositis, or allergic symptoms require urgent evaluation.
- Investigational efficacy and safety are not fully established.
- Keep out of reach of children.
