CAMOBUCOL
Generic Name: CAMOBUCOL
ABSTRACT -
Camobucol (AGI-1067) is a semi-synthetic antioxidant analogue of probucol under investigation for atherosclerosis and cardiovascular disease prevention. It combines anti-oxidative and anti-inflammatory mechanisms, reducing oxidation of LDL cholesterol and modulating inflammatory gene expression in vascular endothelium. Phase III trials (CART1 study) evaluated it for prevention of restenosis after coronary intervention.
DOSAGE -
Oral tablet; 700 mg once daily studied in Phase III trials; taken with food.
SIDE EFFECTS -
- Diarrhoea
- Nausea
- Abdominal discomfort
- QTc prolongation (mild)
- Eosinophilia
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Skin rash
- Urticaria
- Hypersensitivity
- Angioedema (rare)
DRUG INTERACTIONS-
- QT-prolonging drugs — additive QTc prolongation risk
- Cyclosporine absorption may be altered
- Cholesterol-lowering drugs: monitor lipid profile
PREAUTONS–
- Baseline ECG required — QT monitoring during treatment
- Monitor lipid panel and liver function
- Pregnancy safety not established
- Caution in patients with QT abnormalities
WARNINGS –
- QTc prolongation — class effect shared with probucol
- Not approved for cardiovascular prevention — development discontinued after Phase III
- LDL-lowering effect less than modern statins
