CADALIMOGENE IXALENTIVEC

Generic Name: CADALIMOGENE IXALENTIVEC

ABSTRACT -

Cadalimogene ixalentivec (CB-010) is an allogeneic, off-the-shelf CRISPR-engineered CAR-NK or CAR-T cell therapy under investigation for B-cell haematological malignancies. CRISPR editing removes immune rejection signals, enabling use of donor-derived cells without patient-matched manufacturing. Phase I evaluation is ongoing to determine safety and anti-tumour activity.

DOSAGE -

Single or multiple intravenous infusions following lymphodepleting conditioning; cell dose in allogeneic products expressed per fixed dose units per clinical protocol.

SIDE EFFECTS - 

  • Cytokine release syndrome
  • Cytopenias
  • Infections (immunocompromised state)
  • Fatigue
  • Fever

ALLERGIC REACTIONS -

  • These symptoms may indicate an allergic reaction and require immediate medical attention.

    • Infusion hypersensitivity
    • Graft-versus-host disease (GvHD) risk in allogeneic products
    • Anaphylaxis
    • Urticaria

DRUG INTERACTIONS-

  • Lymphodepleting chemotherapy co-administered — interactions with nephrotoxins relevant
  • Avoid live vaccines post-infusion
  • Corticosteroids for CRS management

PREAUTONS–

  • Administered in specialised cellular therapy centre only
  • Infectious disease screening mandatory
  • Monitor for GvHD despite CRISPR editing
  • Long-term follow-up for secondary malignancy

WARNINGS –

  • Life-threatening CRS can occur
  • GvHD risk even with engineered cells
  • Product is allogeneic — off-the-shelf but requires specialised handling
