CROZBACICLIB
Generic Name: CROZBACICLIB
ABSTRACT -
Crozbaciclib is an oral cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor under development for hormone receptor-positive (HR+), HER2-negative breast cancer and other CDK4/6-driven malignancies. By blocking CDK4/6 activity, it arrests tumour cell cycle progression at the G1/S checkpoint. It is being evaluated in combination with endocrine therapies in Phase I/II trials.
DOSAGE -
Oral tablet; dose and schedule (e.g., 21 days on, 7 days off) under determination in Phase I dose-escalation studies.
SIDE EFFECTS-
- Neutropenia

- Diarrhoea
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Skin rash
- Hypersensitivity
- Urticaria
- Angioedema (rare)
DRUG INTERACTIONS-
- Strong CYP3A4 inhibitors increase exposure — avoid or reduce dose
- Strong CYP3A4 inducers reduce efficacy — avoid
- QT-prolonging drugs: monitor ECG
PREAUTONS–
- CBC monitoring before each cycle — dose reduce for neutropenia
- Pulmonary embolism reported — monitor for DVT/PE
- Effective contraception required during and after treatment
- Hepatic impairment — pharmacokinetic data limited
WARNINGS –
