CADONILIMAB
Generic Name: CADONILIMAB
ABSTRACT -
Cadonilimab (AK104) is a bispecific antibody co-targeting PD-1 and CTLA-4 immune checkpoints on T cells, developed by Akeso for solid tumour immunotherapy. Its bispecific design enables simultaneous blockade of two immunosuppressive pathways, potentially offering synergistic anti-tumour efficacy compared to sequential monotherapy. It has received approval in China for recurrent/metastatic cervical cancer and is being evaluated globally.
DOSAGE -
6 mg/kg intravenously once every 3 weeks for approved indication; combination dosing schedules being evaluated in additional tumour types.
SIDE EFFECTS -
- Immune-related adverse events (irAEs): rash, colitis, hepatitis
- Fatigue
- Pruritus
- Nausea
- Hypothyroidism
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Infusion-related reactions
- Severe immune-mediated hypersensitivity
- Anaphylaxis
- Urticaria
DRUG INTERACTIONS-
- Corticosteroids required for irAE management — may reduce anti-tumour efficacy if high-dose
- Avoid immunosuppressive therapies unless treating irAEs
- Live vaccines contraindicated
PREAUTONS–
- Comprehensive baseline assessment: thyroid, LFTs, glucose, autoimmune history
- irAE grading and management per established oncology guidelines
- Effective contraception required
- Withhold or discontinue based on irAE severity
WARNINGS –
- Severe and fatal immune-mediated reactions including pneumonitis, hepatitis, and colitis reported
- Endocrinopathies may be permanent — lifelong hormone replacement required
- Embryo-foetal toxicity — contraindicated in pregnancy
