CADISEGLIATIN
Generic Name: CADISEGLIATIN
ABSTRACT -
Cadisegliatin (HMS5552/RO7009996) is an oral glucokinase activator (GKA) developed for the treatment of type 2 diabetes mellitus. It enhances glucokinase activity in both hepatocytes and pancreatic beta cells, promoting glucose-stimulated insulin secretion and reducing hepatic glucose output. Clinical trials evaluated its glucose-lowering efficacy as monotherapy and in combination with other antidiabetic agents.
DOSAGE -
Investigational oral dosing ranged from 25–75 mg once or twice daily in clinical trials; dose titrated based on glycaemic response and tolerability.
SIDE EFFECTS-
- Hypoglycaemia (dose-dependent)
- Nausea
- Headache
- Dizziness
- Elevated triglycerides
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Skin rash or urticaria
- Facial or throat swelling
- Anaphylaxis (rare)
- Severe pruritus
DRUG INTERACTIONS-
- Additive hypoglycaemic effect with sulfonylureas or insulin
- CYP3A4 inhibitors may increase plasma levels
- Rifampicin and other CYP inducers may reduce efficacy
PREAUTONS–
- Monitor blood glucose regularly to avoid hypoglycaemia
- Use with caution in hepatic impairment
- Not recommended in type 1 diabetes
- Pregnancy and lactation safety not established
WARNINGS –
- Risk of hypoglycaemia especially when combined with secretagogues
- Triglyceride elevations reported; monitor lipid profile
- Discontinue if severe allergic reaction occurs
