COPANLISIB
Generic Name: COPANLISIB
ABSTRACT -
Copanlisib is an intravenous pan-class I phosphoinositide 3-kinase (PI3K) inhibitor with predominant activity against PI3K-alpha and PI3K-delta isoforms. It inhibits tumor cell proliferation and induces apoptosis by blocking PI3K/AKT/mTOR signaling. Approved for relapsed follicular lymphoma and under investigation in other hematologic and solid tumors.
DOSAGE -
60 mg IV infusion over 1 hour on days 1, 8, and 15 of a 28-day cycle.
SIDE EEFECTS -
- Hyperglycemia (transient, infusion-related)
- Hypertension (transient)
- Diarrhea
- Fatigue and asthenia
- Decreased neutrophil count
ALLERGIC REACTIONS -
(These symptoms may indicate an allergic reaction and require medical attention.)
- Infusion-related hypersensitivity
- Skin rash or urticaria
- Anaphylaxis (rare)
- Pruritus or flushing
DRUG INTERACTIONS-
- Strong CYP3A4 inhibitors increase copanlisib exposure
- Strong CYP3A4 inducers (e.g., rifampicin) decrease efficacy
- Avoid concurrent hyperglycemia-inducing agents
PREAUTONS–
- Monitor blood glucose before and 1 hour after each infusion
- Monitor blood pressure pre- and post-infusion
- Risk of serious infections including PCP; prophylaxis recommended
- Non-infectious pneumonitis has been reported
WARNINGS –
- Severe hyperglycemia requiring treatment interruption or dose reduction
- Severe hypertension; antihypertensive pre-treatment may be needed
- Embryo-fetal toxicity — contraception required during therapy
