CEBSULFASE ALFA
Generic Name: CEBSULFASE ALFA
ABSTRACT -
Cebsulfase alfa is a recombinant human lysosomal enzyme replacement therapy (ERT) under development for a rare mucopolysaccharidosis (MPS) disorder characterised by deficiency of a specific sulfatase enzyme, leading to glycosaminoglycan accumulation. Regular intravenous infusions aim to restore enzymatic activity, reduce substrate accumulation, and improve clinical manifestations including organomegaly, skeletal abnormalities, and cardiopulmonary function.
DOSAGE -
Intravenous infusion once weekly or biweekly; dose based on body weight (mg/kg); infusion rate escalated gradually to minimise reactions.
SIDE EFFECTS-
- Infusion-related reactions (fever, chills, urticaria)
- Headache
- Nausea
- Fatigue
- Arthralgia
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Anaphylaxis
- Severe infusion hypersensitivity
- Angioedema
- Bronchospasm
DRUG INTERACTIONS-
- No significant pharmacokinetic drug interactions expected for ERT biologics
- Antihistamine and corticosteroid premedication commonly co-administered
- Immunosuppressants may alter anti-drug antibody response
PREAUTONS–
- Premedication recommended to reduce infusion reactions
- Monitor for antibody development (IgE, IgG)
- Use with caution in respiratory-compromised patients
- Pregnancy registry enrolment encouraged
WARNINGS –
- Life-threatening anaphylaxis may occur — have resuscitation support available
- Infusion must be administered in a clinical setting with monitoring
- Immunogenicity may reduce long-term efficacy
