AMCASERTIB
Generic Name: AMCASERTIB
ABSTRACT -
Amcasertib, also known as BBI503, is an investigational orally active small molecule studied for anticancer activity. It has been described as a cancer-stemness kinase inhibitor that affects pathways associated with NANOG, CD133, tumor-cell survival, self-renewal, and differentiation.
Clinical studies have evaluated amcasertib in advanced solid tumors and hepatobiliary cancers. It remains investigational and is not approved for general clinical use.
DOSAGE -
Dosage should be determined only by a qualified oncologist within a clinical-trial protocol.
No approved dosing regimen exists for Amcasertib.
Investigational studies have used oral daily dosing schedules in continuous cycles, with dose and duration defined by protocol.
Do not use outside approved research, compassionate-use, or regulatory-authorized settings.
SIDE EFFECTS–
Possible side effects reported or expected in oncology studies include:
- Nausea, vomiting, or diarrhea
- Loss of appetite
- Fatigue or weakness
- Abdominal discomfort
- Rash
- Laboratory abnormalities in liver function or blood counts
- Dehydration secondary to gastrointestinal toxicity
- Unknown severe adverse events due to investigational status
ALLRGIC REACTIONS–
Hypersensitivity to amcasertib or formulation components is possible. Specific cross-reactivity information is not established.
Symptoms may include:
- Skin rash or hives
- Itching or swelling of the face, lips, tongue, or throat
- Difficulty breathing or chest tightness
- Severe dizziness or fainting
- Unexpected widespread redness, blistering, or peeling skin
Important: Patients who develop an allergic reaction to Amcasertib should seek immediate medical attention and inform all future healthcare providers of the allergy.
DRUG INTERACTIONS–
Drug-interaction data are limited. Caution is needed with:
- Other investigational oncology agents
- Cytotoxic chemotherapy or targeted therapies that may add gastrointestinal, hepatic, or marrow toxicity
- Strong metabolic enzyme inhibitors or inducers if allowed by protocol
- Anticoagulants or antiplatelet drugs in patients with thrombocytopenia risk
- Herbal products used by cancer patients, especially St. John's wort
Patients should inform healthcare providers about all prescription medicines, over-the-counter medicines, herbal products, and supplements they are using.
PREAUTIONS–
- Use only under oncology trial supervision.
- Monitor blood counts, liver function, hydration status, and gastrointestinal symptoms.
- Assess performance status and organ function before use.
- Pregnancy should be avoided; embryo-fetal risk is unknown but possible for anticancer agents.
- Report severe diarrhea, persistent vomiting, fever, bleeding, or jaundice immediately.
WARNINGS –
- Amcasertib is investigational and not approved as standard cancer therapy.
- Do not combine with other anticancer drugs unless directed by a clinical-trial protocol.
- Severe dehydration or organ toxicity can occur with uncontrolled gastrointestinal adverse effects.
- Patients should not stop or substitute approved cancer therapy without oncology advice.
- Keep out of reach of children.
