ARZOXIFENE
Generic Name: ARZOXIFENE
ABSTRACT -
Arzoxifene is a selective estrogen receptor modulator investigated for osteoporosis, breast cancer prevention, and hormone-responsive malignancies. It was designed to provide estrogen-antagonist effects in breast and endometrium while preserving beneficial bone or lipid effects. Development has remained investigational rather than routine clinical use.
DOSAGE -
- No approved routine dosage regimen is established.
- Clinical studies have evaluated oral once-daily dosing, including 20 mg daily in some programs, but this should be considered investigational.
- Do not substitute for approved SERMs such as tamoxifen or raloxifene without specialist direction.
- Patients require individualized assessment of menopausal status, cancer risk, bone risk, and thromboembolism risk.
SIDE EFFECTS–
- Hot flushes, sweating, leg cramps, arthralgia, or musculoskeletal pain
- Peripheral edema, nausea, abdominal discomfort, or headache
- Vaginal discharge, bleeding, or gynecologic symptoms
- Venous thromboembolism, stroke risk, or cardiovascular events in susceptible patients
- Rash or hypersensitivity reactions
ALLERGIC REACTIONS -
- Allergy may include rash, hives, facial swelling, breathing difficulty, severe dizziness, or anaphylaxis.
- Unexplained vaginal bleeding or severe allergic symptoms require urgent evaluation.
DRUG INTERACTIONS-
- Estrogens, hormone therapy, tamoxifen/raloxifene, anticoagulants, warfarin, and strong metabolic inhibitors/inducers require review.
- Immobilization, surgery, or cancer therapy can increase thrombosis risk.
- Avoid combining with other SERMs unless explicitly directed.
PREAUTIONS–
- Assess history of VTE, stroke, cardiovascular disease, liver disease, abnormal uterine bleeding, pregnancy status, and cancer risk.
- Monitor for leg swelling, chest pain, dyspnea, neurologic symptoms, and gynecologic symptoms.
- Discontinue before prolonged immobilization or surgery if directed by protocol.
- Not intended for use in pregnancy.
WARNINGS –
- SERMs can increase risk of serious blood clots and stroke in susceptible patients.
- Investigational status means routine benefit-risk is not established.
- Seek urgent care for unilateral leg swelling, chest pain, sudden shortness of breath, severe headache, or neurologic deficits.
- Keep out of reach of children.
