Generic Name: CAMIBIRSTAT
ABSTRACT -
Camibirstat is an oral, selective inhibitor of heparanase (HPSE), an enzyme that cleaves heparan sulfate proteoglycans in the extracellular matrix, promoting tumour invasion, angiogenesis, and metastasis. By blocking heparanase, camibirstat aims to reduce tumour dissemination and remodel the tumour microenvironment. It is under evaluation in Phase I/II trials for solid tumours including multiple myeloma and solid cancers.
DOSAGE -
Oral tablet; dose escalation ongoing in Phase I studies; investigated as monotherapy and in combination with standard-of-care agents.
SIDE EFFECTS -
- Nausea
- Fatigue
- Diarrhoea
- Headache
- Elevated liver enzymes
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Skin rash
- Urticaria
- Hypersensitivity
- Angioedema (rare)
DRUG INTERACTIONS-
- No major CYP interactions identified at this stage
- Additive anti-tumour effect with bortezomib in myeloma under study
- Monitor with anticoagulants — heparan sulfate pathway interactions
PREAUTONS–
- LFT monitoring during treatment
- Bone marrow function monitoring if myeloma patient
- Effective contraception required
- Paediatric safety data not available
WARNINGS –
- Investigational agent — full adverse event profile under characterisation
- Hepatotoxicity not excluded — regular LFT monitoring mandatory
- Embryo-foetal toxicity cannot be excluded
