CAMLIPIXANT
Generic Name: CAMLIPIXANT
ABSTRACT -
Camlipixant (BAY 1817080) is a highly selective oral antagonist of the P2X3 purinoceptor, developed by Bayer for the treatment of refractory chronic cough (RCC). P2X3 receptors on sensory afferent nerves mediate cough hypersensitivity responses; antagonism of these receptors reduces cough frequency without significant dysgeusia. Phase II trials demonstrated meaningful reductions in 24-hour cough frequency compared to placebo.
DOSAGE -
Oral tablet; investigational doses of 50–200 mg twice daily evaluated in Phase II trials; optimal dose being refined in Phase III studies.
SIDE EFFECTS-
- Taste disturbance (dysgeusia) — less frequent than with gefapixant
- Headache
- Nasopharyngitis
- Nausea
- Dizziness
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Skin rash
- Hypersensitivity reactions
- Angioedema (rare)
- Urticaria
DRUG INTERACTIONS-
- CYP3A4 inhibitors may increase camlipixant exposure
- No significant interaction with commonly used antitussives
- Monitor with potent CYP inducers
PREAUTONS–
- Confirm refractory chronic cough diagnosis before initiating
- Use with caution in severe hepatic impairment
- Not studied in paediatric populations
- Pregnancy safety not established
WARNINGS –
- Dysgeusia, although less than P2X3 antagonist class predecessors, may occur
- Patients should report persistent or worsening taste changes
- Avoid in known hypersensitivity to the drug or excipients
