CALPURBATUG
Generic Name: CALPURBATUG
ABSTRACT -
Calpurbatug is an investigational antibody-drug conjugate (ADC) targeting prostate-specific membrane antigen (PSMA), designed for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The antibody component selectively binds PSMA-expressing tumour cells, delivering a cytotoxic payload intracellularly to induce apoptosis. It is being evaluated in Phase I/II trials for patients with progressive disease following standard therapies.
DOSAGE -
Administered intravenously; dosing schedule and recommended dose being determined in ongoing Phase I dose-escalation studies.
SIDE EFFECTS-
- Fatigue
- Nausea and vomiting
- Peripheral neuropathy
- Haematological toxicity (neutropenia, thrombocytopenia)
- Alopecia
ALLERGIC REACTIONS -
These symptoms may indicate an allergic reaction and require immediate medical attention.
- Infusion-related reactions (chills, fever, hypotension)
- Anaphylaxis
- Urticaria
- Bronchospasm
DRUG INTERACTIONS-
- Avoid concurrent use with other myelosuppressive agents
- Caution with CYP3A4 inhibitors affecting payload metabolism
- Monitor with anticoagulants due to thrombocytopenia risk
PREAUTONS–
- Premedicate to reduce infusion reactions
- Monitor CBC prior to each cycle
- Use contraception during and for 6 months post-treatment
- Renal dose adjustment may be required
WARNINGS –
- Severe myelosuppression reported; dose reductions may be necessary
- Peripheral neuropathy may be cumulative and irreversible
- Embryo-foetal toxicity — avoid in pregnancy
